Evaluación in vitro de la actividad antitumoral de extractos naturales sobre células positivas para el virus del papiloma humano

Resumen

El cáncer oral es una patología tumoral de comportamiento agresivo que representa de 2-5% de las neoplasias diagnosticadas produciendo una elevada tasa de morbi-mortalidad a nivel mundial. En Colombia es considerada prioridad en salud pública, con una mayor incidencia en hombres. Los factores de riesgo: consumo de alcohol, tabaco e infección por el virus del papiloma humano. Los tratamientos convencionales como cirugía, quimioterapia y radioterapia, producen efectos adversos. Actualmente, no existen tratamientos específicos contra la infección oral producida por el virus, y las vacunas no han demostrado efectividad. Por tal razón, existe la necesidad de encontrar nuevos tratamientos para reducir la infección de manera significativa. Evaluar el efecto de los extractos naturales obtenidos a partir de; Trametes versicolor, Kudzu root, Schizandra, Dodder seed, Wolfberry, Solanum lycopersicum frente a células infectadas con el virus. Extractos preparados en dimetilsulfóxido evaluados en la línea celular HeLa de queratinocitos cervicales infectados con VPH18. Se realizaron ensayos de citotoxicidad con tres réplicas experimentales y tres biológicas por el método de resazurina. Se evaluó la expresión de los oncogenes E6/E7 y p53 por RT-PCR y expresión de la proteína p53 y pRb por Western blot. De acuerdo a la evaluación de la citotoxicidad los extractos indujeron muerte celular a las concentraciones evaluadas (10005000µg/mL). Con respecto a la expresión de las oncoproteínas E6/E7 se demostró que los extractos de Trametes versicolor (0,1, 1µg/mL), Wolfberry (0,1, 1 y 10µg/mL) inhiben la expresión de E6, y a todas las concentraciones evaluadas inhiben la expresión E7, mientras Té verde solo a concentración de 10µg/mL indujo efectos sobre estas oncoproteínas, se demostró que los extractos de Trametes versicolor y Wolfberry (0,1, 1µg/mL) aumentaron de manera significativa la expresión de p53 y la producción de esta proteína mediante Western blot (p>0,05) diferente al control positivo. Ninguno presentó cambios en la expresión de la Fosforilación de la proteína retinoblastoma. Conclusiones: los extractos de Trametes versicolor y Wolfberry poseen efectos inhibitorios de la expresión de las oncoproteínas E6/E7, al igual que un aumento de la producción de p53, demostrando que estos tratamientos podrían tener efectos antivirales frente a VPH18.

Abstract

Oral cancer is a tumoral pathology with an aggressive behaviour which represents 2% to 5% of diagnosed neoplasias producing an elevated rate of morbi-mortality worldwide. It is considered a public health priority with higher prevalence among men and risk factors such as: alcohol consumption, tobacco and infection due to the human papillomavirus. Conventional treatments such as surgery, chemotherapy and radiotherapy produce adverse effects. Currently there are no specific treatments against infection produced by the virus and vaccines have not shown any effectiveness. This creates the necessity of finding new treatments which significantly reduce the infection. To evaluate the effect of natural extracts obtained from Trametes versicolor, Kudzu root, Schizandra, Dodder seed, Wolfberry, Solanum lycopersicum on cells infected by the virus. The extracts were prepared in dimethyl sulfoxide evaluated in the cellular line HeLa of cervical keratinocytes infected with HPV18. Cytotoxicity trials with three experimental replicas and three biological with the resazurin method were carried out. The expression of E6/E7 and p53 oncogenes by RT-PCR was evaluated and the expression of p53 and pRb protein by Western blot. Cytotoxicity assessment yielded that the extracts induced cellular death with the evaluated concentrations (1000 µg/mL -5000 µg/mL). The Trametes versicolor (0,1, 1µg/mL), Wolfberry (0,1, 1 and 10µg/mL) inhibit the expression of E6, all evaluated concentrations inhibit the expression of E7 and Té verde only induced effects on the oncoproteins with a concentration of 10µg/mL. Trametes versicolor y Wolfberry (0,1, 1µg/mL) extracts increased significantly the expression of p53 and its production by Western blot (p>0.05) different to the positive control. None presented changes in the expression of the phosphorylation of the retinoblastoma protein. Trametes versicolor and Wolfberry extracts possess inhibitory effects on the expression of E6/E7 oncoproteins as well as a production increment of p53, demonstrating that these treatments could have anti-viral effects against HPV18.
